Isolation and Identification of Novel Penicillium and Aspergillus Species Isolated from Soil in Korea

Two fungal strains, designated as KNUF-25-MP1 and KNUF-25-TY8, were isolated from soil samples collected in Daegu-si and Geochang-gun, Korea, respectively. To observe fungal characteristics, strains were cultured on potato dextrose agar (PDA), Czapek yeast autolysate agar (CYA), yeast extract sucrose agar (YES), malt extract agar (MEA), creatine sucrose agar (CREA), dichloran 18% glycerol agar (DG18), oatmeal agar (OA), synthetic nutrient agar (SNA). Phylogenetic analysis was performed using sequences of internal transcribed spacer (ITS) regions, β-tubulin (BenA), calmodulin (CaM), RNA polymerase II subunit (RPB2), pre-rRNA processing protein (Tsr1) genes. BLAST results of ITS regions, BenA, CaM, RPB2 gene sequences of KNUF-25-MP1 showed 99.5, 95.5, 95.5, 99.8% similarities, respectively, to closely related species Penicillium fructuariae-cellae ITEM18276T and formed distinct cluster from other Penicillium species in phylogenetic tree. Morphologically, KNUF-25-MP1 was distinguished by cultural characteristics on CYA, YES, MEA, CREA, as well as by producing larger conidia than P. fructuariae-cellae ITEM18276T (3.0–4.5 µm vs. 2.0–3.5 µm). In the case of KNUF-25-TY8, BLAST results of ITS regions, BenA, CaM, RPB2, Tsr1 gene sequences showed 93.2, 87.5, 87.1, 91.3, 90.8% similarities, respectively, to closely related species Aspergillus doliiformis GZUIFR21.885T and formed distinct cluster from other Aspergillus species in phylogenetic tree. Morphologically, KNUF-25-TY8 was distinguished by cultural characteristics on PDA, OA, MEA, as well as by producing smaller conidia than A. doliiformis GZUIFR21.885T (2.5–3.5 × 2.0–2.7 µm vs. 3.0–5.0 × 2.5–4.5 µm). Based on morphological and phylogenetic analyses, we proposed novel Penicillium and Aspergillus species isolated from soil in Korea.
